A Sensitive and Reproducible Assay for the Quantitation of the in Vitro Antibody Response to Sheep Red Blood Cells and the Effect of Thymocytes, Endotoxin, and Phytohemagglutinin on this Response

Abstrakt: A new and sensitive assay procedure is described for the study of the in vitro production of antibody by lymphoid cells. The in vitro culture technique and the hemolysin assay procedure enable quantitation of the effects of various cell types, chemicals, hormones, or cell products on antibody stimulation. The method allows large numbers of variables to be assessed in replicate. The addition of either thymocytes or endotoxin is necessary to obtain maximum antibody production, whereas, only at high concentrations does PHA affect this response. In vitro antibody production by spleen cells is more dependent upon the visible condition of the donor animal's thymus than on the addition of special enhancing sera or serum fractions. Implications of these findings are discussed.
