Isolation and Identification of human bone mesenchymal stem cells in fibrous dysplasia of bone in vitro.

Abstrakt: PURPOSE: To investigate a method of isolation and identification of bone mesenchymal stem cells from patients with fibrous dysplasia of bone in vitro and lays the foundation for the study of the etiology and pathogenesis of fibrous dysplasia. METHODS: The fresh bones of patients with FD were obtained.The samples were cut and washed into dish so as to acquire the bone mesenchymal stem cells (MSC). After cell culture, their morphology was observed,cell growth curve was drawn,and cell surface marker and osteogenic differentiation potency were detected. RESULTS: With the increasing number of MSC passage,the MSC morphology trended to be fusiform shape. 99% of the cells expressed CD44 and HLA-ABC, but no expressed CD34. The cells were positive for alkaline phosphatase staining and calcium nodules after being induced. CONCLUSION: MSCs can be successfully obtained from the fresh specimens of FD and can be induced for osteogenesis in vitro. The acquired MSCs can meet the subsequent experimental request.
