105. Clinical Relevance of Circular RNA circ_0005402 Expression in Multiple Sclerosis in a Cohort of Egyptian Patients.

Abstrakt: Dysregulated profiles of certain circular RNAs (specifically ANXA2 circular RNAs circ_0005402), have recently been demonstrated in patients with Multiple Sclerosis (MS). Nevertheless, despite that circular RNA circ_0005402 has been proven to be differentially expressed in MS with high sensitivity (90.0%) and moderate specificity (60.0%), a huge gap of knowledge still exists regarding its exact role in the immunopathogenesis of MS. Noteworthy, ANXA2 is a target of miR-155, the latter has been positively associated with MS disease severity. Furthermore, Bioinformatics' analysis of the sequence of circ_0005402 revealed a binding site for miR-766, an anti-inflammatory microRNA, in addition to five binding sites for RNA-binding proteins, including AGO2, EIF4A3, LIN28A, IGF2BP1, and PTB. Interestingly, LIN28 has an imperative role in promoting axon regeneration in the central nervous system. This study was conducted on 30 confirmed MS patients who had never received any disease modifying therapy. Circular RNA circ_0005402 expression using reverse transcription real-time polymerase chain reaction was correlated with different parameters. No significant correlation was observed between circ_0005402 values and age (P = 0.719), age of onset of MS (P = 0.279), disease duration (P = 0.171), time interval between the first demyelinating event and the first relapse (P = 0.142), and Expanded Disability Status Scale (P = 0.129). Furthermore, no significant association was observed between circ_0005402 values and sex (P = 0.471), type of MS (P = 0.347), and presence of family history (P = 0.294). A positive association was found only between experiencing motor manifestations in the first demyelinating event and circ_0005402 values (P = 0.031). From this study, we concluded that circular RNA circ_0005402 is only positively associated with experiencing motor manifestations in the first demyelinating event, thus it cannot be used as a prognostic marker; nevertheless, this opens the door for future research to investigate the exact pathological role circular RNA circ_0005402 plays in MS.
