Callus production from isolated microspore culture of peanut (Arachis hypogaea L. ).

Abstrakt: The relevance of pollen development stages, shape of bud and anther was discussed, and the effect of genotypes of isolated microspore, different kinds of liquid medium as well as additional hormone concentration on the callus was studied. The results showed that, when the longitudinal diameter of bud were 2. 48 ~2. 96mm, 3. 26 ~4. 16mm and 5. 38 ~9. 9mm, development stages of pollen were mostly at tetrad, mononuclear period and uninu-cleate respectively. At late - uninucleate, the induction rate was the highest, at 9. 38%. Six varieties of peanut were used in this experiment, the average embryo yield of Silihong was the highest, with 0. 1 embryo per bud (MS medium) and 0. 05 embryo per bud ( NLN medium) . On MS medium of 4mg/L 2, 4 - D, 3mg/L 6 - BA and 0. 3 mg/L NAA, with 2. 5 x 105/mL concentration of microspore, the induction rate of Silihong was the highest at 12.5%. [ABSTRACT FROM AUTHOR]
