K-strophanthoside(H) was administered rectally to 96 guinea-pigs at doses of 250, 500 and 1000 μg/kg. The guinea-pigs were sacrified after 30 min, 1, 5 and 15 h. Drug levels were determined in the heart, liver, kidneys, skeletal muscle, carcass, small intestine wall and contents, large colon wall and contents, rectum wall and contents, blood, bile and urine. The highest concentrations were found in the intestine contents, rectum walls, bile and urine. The amount excreted with urine was less than 1% at the 30th min and 1st h, about 9% at the 5th h and 17-18% at the 15th h. Concentrations in the guinea-pig organs, tissues and fluids proved to be closely proportionate to the doses administered, as did the amounts of the drug absorbed and those excreted with urine. The enteral absorption of K-strophanthoside amounted to 65.57% of the dose administered rectally over a 5 h period in anesthetized guinea-pigs. In conscious guinea-pigs the enteral absorption calculated from urinary excretion data after rectal and i.v. administration was around 55% over a 15 h period. [ABSTRACT FROM AUTHOR] |
