This paper presents results of a combined study on morphological, physiological and biochemical peculiarities of Arthrobacter globiformis 193 cells immobilized in polyacrylamide during repeated batchwise transformations of hydrocortisone in buffer. The cells, entrapped in gel under optimal conditions, had a specific Δ-dehydrogenase activity equal to the activity of free cells. The half life of immobilized cells was 140 days (160 transformations) while the net operation time was 20 days. The incubation of granules in a nutrient medium favoured cell propagation within granules and increased the enzymic activity, stability and efficiency of the immobilized system. After 6 months (200 transformations), granules incubated once and twice in the nutrient medium retained 72% and 92% respectively of the activity of newly made granules. The decrease in the activity was due to cell lysis which increased with time towards the granule centre. In the subsurface layer, the intact cells remained viable for a long time and so stabilized the enzymic activity of the immobilized system. [ABSTRACT FROM AUTHOR] |
