Abstrakt: The article discusses generating knockout and tagged human cell lines by using zinc finger nucleaze (ZFN), an alternative way of developing oncology cell lines. ZFNs are reported to be also useful in inserting reporter genes into the target organism or cell line. The authors discuss cell culture, nucleofections, and the ELISA modified kit from R&D Systems. They include charts that show typical tubulin bundle formation, ZFNs binding to the target site, and the morphological similarity between wild type and red fluorescent protein (RFP)-tagged lines.
