Protein engineering of Acidovorax facilis 72W nitrilase for bioprocess development.

Abstrakt: The article presents a study on the use of protein engineering in increasing activity of the Acidovorax facilis (A. facilis) 72W. The A. facilis 72W nitrilase converts 3-hydroxynitriles to 3-hydroxycarboxylic acid which can be used for the preparation of polyesters. The Genomic DNA was prepared from A. facilis 72W using the Puregene DNA kit while saturation mutagenesis was done using degenerate oligonucleotides and the QuikChange Site-Directed Mutagenesis Kit.
