A Morphologically Recognizable Marker for Scanning Immunoelectron Microscopy.

Abstrakt: Antibody-induced redistribution of virus-induced surface antigens (VSA) at the surface of MDCK cells infected with influenza B virus has been studied by scanning immunoelectron and immunofluorescence microscopy. Redistribution was not observed on the contact-inhibited monolayer cells, whereas it was evident on the dispersed suspension cells. After the addition of antl-VSA antibody, almost all of the suspended cells showed patchy distribution and a subsequent polar distribution (cap) of VSA. This capping of VSA was completely inhibited by cytochalasin B. Scanning Immunoelectron microscopy using bacteriophage T4 as a visual marker (T4-IgG labeling) dramatically demonstrated such redistribution of VSA with a higher sensitivity.
