An Ultrastructural Study of Reparative Dentinogenesis in the Rat Incisor after Colchicine Administration.

Abstrakt: Reparative dentinogenesis after administration of a large dose of colchicine was studied in rat incisors. Odontoblasts and the adjacent pulp tissue were destroyed soon after colchicine administration. At 6 days, spindle-shaped cells surrounded by a fibrous matrix appeared at the interface between the necrotic matrix and the normal pulp. Between 8–10 days, short columnar odontoblast-like cells had differentiated adjacent to a newly developing pre-dentin-like matrix. By 10–12 days, new tubular dentin-like structures had been formed in contact with the non-mineralized collagenous matrix. Various kinds of membrane-bounded extracellular vesicles were distributed between the collagen fibrils in the predentinal matrix produced by the new odontoblasts. Their morphological characteristics corresponded with true matrix vesicles. From these observations, it is suggested that the initial calcification process in reparative dentinogenesis after colchicine administration is essentially similar to the initial calcification process of normal dentin, namely by matrix vesicular calcification.
