The arrangement of chromosomal fibers was examined for the root meristematic cells of (broad beans) by SEM. The root tips were freeze-fractured in liquid nitrogen. After being thawed in a DMSO solution, they were incubated in a buffer solution containing NaCl, and then fixed with glutaraldehyde. Parallel longitudinal arrangement of fibers was noticed in chromatids by the treatment with NaCl. The extraction of histones with Nacl may cause stretching of chromosomal fibers, which might have been folded back and forth repeatedly for some distance in a chromatid. [ABSTRACT FROM PUBLISHER] |
