To compare the nephrotoxic potentials of dibekacin, gentamicin and tobramycin in rats at doses similar to those employed clinically, we gave those drugs at 1, 2 and 4 times the recommended human dose on a weight basis for 14 days. Urinary protein excretion, urinary osmolality, endogenous creatinine clearance and an elaborate discrete pathological grading scale were employed. Dose related changes in urine osmolality occurred, while urine protein excretion and creatinine clearance were not consistently affected. The regimens also caused modest dose related changes in renal tissue. No clear differences in toxicity were identified at these doses. We conclude that at low doses, these drugs are similar in their proclivity to cause renal damage. [ABSTRACT FROM PUBLISHER] |
