The application of microbore liquid chromatography/mass spectrometry to the routine analysis of a drug in plasma.

Abstrakt: We report on a validated assay of a new drug candidate using a 1 mm inner diameter (i.d.) analytical column and semi-automated sample preparation with solid phase extraction in the 96-well plate format. The method was used during a phase-l study in which the results had to be delivered on 102 days within 8 hours. Compared to the original assay using a 2 mm i. d. column, the performance of the new assay is increased. The decrease of the inner diameter from 2 to 1 mm (and the adjustment of the chromatographic conditions) leads to shorter analysis time and enhanced sensitivity without compromising the robutness of the assay. The lifetime of the analytical 1 mm i. d. column was 2500 injections, the one of the guard column about 1300 injections. [ABSTRACT FROM AUTHOR]
