Demonstrates that the hypoxanthine phosphoribosyl-transferase (HPRT) gene, which is mutated in humans with Lesch-Nyhan disease can be corrected by homologous recombination in colony-forming cells. Survival of corrected colonies in selective medium after electroporation of correcting DNA into unfractionated mouse bone marrow cells. |
