The article discusses improvements to processes involving the salt fractionation of animal serums in order to prepare for immunofluorescence studies. The tendency for dye to attach to the albumin within serum fractions containing appreciable amounts of albumin upon labeling with fluorescein isothiocyanate is noted, mentioning that the attachment of dye to albumin is due to its affinity for the isothiocyanate group. The inability to calculate a meaningful fluorescein-protein (F/P) ratio in situations in which a conjugate contains labeled nonantibody proteins such as albumin in significant or variable amounts is also noted. |
