Aspirin, a non steroidal anti-inflammatory drug, is associated with side effects including intestinal discomfort and increased blood loss. Recently, novel drug delivery systems have been developed to deliver drugs with minimal side effects while maintaining the efficacy of the drug. We have formulated a soybean oil, water and polysorbate 80 nano emulsion preparation for aspirin utilizing microfluidization for comparison with an aspirin suspension as it relates to their respective anti inflammatory properties in the croton oil-induced ear edema CD-1 mouse model. Measurements of anti-inflammatory properties included ear lobe thickness as well as the accumulation of the pro inflammatory cytokines levels of IL-1á, TNFá and IL-6 in ear lobe homogenates. The results showed that nanoemulsion preparation of aspirin was a homogenous population (∼96%) of particles with an average particle size of 118nm. In addition, the reduction in inflammation as measured by changes in ear lobe thickness was approximately 2X times greater for the nanoemulsion compared to the suspensions of aspirin. Furthermore, a statistically significant reduction (P<0.05) was seen for the ear lobe homogenate cytokine levels which also correlated with the difference in ear lobe thickness. In summary our study suggests that a nanoemulsion preparation of aspirin improves the anti-inflammatory property of aspirin in an ear edema mouse model. [ABSTRACT FROM AUTHOR] |
