Purification of IgG from serum by automation with pre-filled reagent cartridges and a magnetic separation robot.

Abstrakt: PSS Bio has recently developed an automated His-tagged protein purification system using pre-filled reagent cartridges and a magnetic filtration (Magtration®) robot. In this report, we demonstrate a new application of this technology to purify IgG from serum. Up to 12 samples can be purified in parallel by automation on a Magtration® 12GC robot using pre-filled reagent cartridges and protein A-conjugated magnetic beads. The automated protocol has been optimized and preprogrammed for the purification. The magnetic beads are automatically regenerated after a purification using the same cartridge and are ready for reuse in additional lgG purifications. At least 340 µg of IgG can be purified from a 30 µl human serum sample. A total of about 4 mg of human IgG can be purified by processing 12 samples simultaneously with less than one hour of processing time. This protocol can be extended to purify IgG from other samples such as mouse monoclonal ascites fluid, and rabbit and guinea pig sera. With the Magtration® robotic system, affinity purification of IgG and other target proteins can be fully automated with walk-away ease of use. [ABSTRACT FROM AUTHOR]
