STUDY OF ALLELIC VARIANTS' STRACTURE OF LOCI Du281 IN Darevskia dahli (LACERTIDAE) PARTHENOGENETIC LIZARDS POPULATIONS.

Abstrakt: The genesis of mini- and microsatellite loci, which is under extensive study in humans and some other bisexual species, have been virtually overlooked in clonal mode of reproduction. In present study we use PCR- analyze to examine allelic polymorphism of two loci of D. dahli - Du 215 and Du 281- containing (GATA)n microsatellite clusters. Different levels of intrapopuliation and interpopuliation variability of these loci were found. Du 215 showed the highest polymorphism - 8 allelic variants. Three alleles were found for Du 281. Earlier, we used PCR-analysis for homologous loci in two presumptive parental bisexual species D. mixta and D. portschinski. To understand the molecular basis of microsatellite variability it is important to know detailed structure of microsatellite alleles and their possible turn over. PCR products produced from polymorphic microsatellite loci of D. dahli - Du 281 were also sequenced and allelic variants of these loci were revealed. We found that allelic differences of this locus were caused by variation in a number of tandem repeats in microsatellites clusters and point mutations in the flanking regions. This information is significant for understanding of genetic variability in parthenogenetic lizard species and for study of phylogenetic relationship in unisexual and related bisexual species of genus Darevskia.
