The article examines a sensitive method for the analysis of urinary ribonucleosides including affinity chromatography purification prior to liquid chromatography-mass spectrometry analysis using a micrOTOF ESI-oa-TOF-MS system. Reversed-phase chromatographic separation of the prepurified nucleosides was performed using an Agilent 1100 binary gradient pump HPLC system with photodiode array detection and a XTerra C18 column. The study shows the possibility to generate structural hypothesis for urinary nucleosides based on accurate and true isotopic pattern in combination with database search. |
