The highly contagious avian infectious bronchitis virus (IBV) typically affects the respiratory, renal, and reproductive systems in chickens. It has a worldwide distribution and a high morbidity rate which may reach 100% inducing severe economic losses. The present study was undertaken to investigate the seroprevalence and molecular detection of IBV in backyard and commercial poultry in Assam, India for five years (2019 to 2024). Sera samples (n=2266) were collected and assayed by an IBV antibody detection indirect ELISA. Serological analysis showed 20.154% and 22.881% seroprevalence of IBV in backyard and commercial poultry flocks respectively. Additionally, 121 suspected tissue samples were investigated and analyzed using reverse transcriptase polymerase chain reaction (RT-PCR) targeting a 656 bp S' gene of IBV Seventy-four (61.157%) samples were positive by RT-PCR. Our study demonstrated the prevalence of IBV in backyard and commercial poultry of the region. Therefore, monitoring and typing of IBV are needed to determine its impact on the poultry industry as well to choose adequate vaccine strains for strategic control of the disease.
