Development and Characterization of Paclitaxel-Loaded MF59 Nano-emulsion for Breast Cancer Therapy.

Abstrakt: This study aimed to evaluate the potential of MF59 nano-emulsion as a carrier for the sustained release of paclitaxel (PTX) and its effects on the human breast cancer cell line Michigan Cancer Foundation (MCF7) and human dermal fibroblasts (HDF). Due to PTX's low solubility and toxicity, a safer and more effective delivery system is needed for breast cancer therapy. A paclitaxel-loaded MF59 nanoemulsion (PTX-MF59) was formulated using the homogenization method and characterized for particle size, polydispersity index (PDI), zeta potential, encapsulation efficiency (EE), drug loading (DL), in vitro release profile, and stability using dynamic light scattering (DLS), scanning electron microscopy (SEM), Fourier-transform infrared (FT-IR), and ultraviolet–visible (UV–VIS) techniques. Cytotoxicity was assessed in MCF-7 breast cancer cells and HDF normal fibroblasts using the MTT assay to determine inhibitory concentration (IC50) values. The nano-emulsion had a mean particle size of 119.7 ± 37.30 nm with a PDI of 0.12. Encapsulation efficiency (EE) and drug loading (DL) were 88% and 4.4%, respectively. In vitro release showed an initial burst release of 42% over 2 days, followed by sustained release up to 26 days. The inhibitory concentration (IC50) values of free paclitaxel, MF59 and PTX-MF59 were 10.59, 44.46, 20.15 µg/mL for MCF-7 cells and 12.06, 44.65, 30.17 µg/mL for HDF cells, respectively. The formulated paclitaxel-loaded MF59 nano-emulsion exhibited favorable characteristics and significantly higher cytotoxicity in MCF-7 cells compared to normal cells, indicating its potential as a safer delivery system for breast cancer therapy.
