Researchers at the Karlsruhe Institute of Technology (KIT) in Germany have developed a new method for quantifying lipids in lipid nanoparticles (LNPs), which are potential delivery systems for nucleic acid payloads. The method, called RP-CAD, uses reversed-phase charged aerosol detection and was found to be effective in characterizing LNPs and their processes. The study also identified deviations in lipid content and lipid loss during LNP processing, which were attributed to lipid stock solution preparation and small-scale dialysis. This research lays the foundation for using RP-CAD in lipid quantification throughout LNP processing and suggests its potential applicability to other LNPs and process parameter studies.
