Effect of circ_0007059 targeting miR-1303 on malignant biological behavior of LNCaP cells in prostate cancer.

Abstrakt: Objective To investigate the effect of circ_;0007059 on malignant biological behavior of prostate cancer LNCaP cells. Methods The cancer tissue and paracancerous tissue specimens of 41 prostate cancer patients in our hospital from May 2017 to 2020 were selected. In vitro culture of prostate cancer LNCaP cells, plasmid transfection was used to overexpress/knockdown circ_;0007059 expression, inhibit miR-1303 expression, and up-regulate circ_;0007059 and miR-1303 expression, respectively. After transfection, circ_;0007059 and miR-1303 expression, cell proliferation, migration, invasion, Ki-67, MMP-2, MMP-9 protein expression were detected, and the targeting relationship between circ_;0007059 and miR-1303 was verified. Results The expression of circ_;0007059 in prostate cancer tissue was decreased, and the expression of miR- 1303 was increased (P < 0. 05). After circ_;0007059 overexpression or inhibition of miR-1303 expression, the LNCaP cells viability, migration and invasion ability and Ki--57, MMP-2 and MMP-9 protein levels was decreased. circ_;0007059 targets and regulates miR-1303 ; up-regulation of miR-1303 reversed the effects of circ_;0007059 overexpression on LNCaP cells. Conclusions Overexpression of circ _;0007059 can inhibit the malignant biological behavior of LNCaP cells, and the mechanism may be related to the targeted down-regulation of miR-1303.
