HPLC method was developed for the simultaneous determination of 10 flavonoid compounds in different parts of sweet sorghum, and the antioxidant activity of total flavonoid extracts was evaluated. The total flavonoid extracts were prepared from mature seeds, stems, and the whole plant of sweet sorghum cultivar Longtianliang No. 2. HPLC was employed to simultaneously determine the content of catechin, apigenin, quercetin, rutin, kaempferol, hesperidin, naringin, hyperoside, luteolin, and maysin content in different parts of total flavonoid extracts. The results showed that the 10 flavonoid compounds exhibited good linearity with peak area within their respective linear ranges, with R² ranging from 0.998 4 to 0.999 9. The average recovery rates were between 95.0% and 105.8%, and the RSD was in the range of 1.03% to 2.86%. The antioxidant activity assays revealed that the total flavonoid extracts from different parts of sweet sorghum had a certain scavenging capacity against hydroxyl radical, superoxide anions radical, DPPH free radical, and ABTS+ free radical, and the scavenging rate at different concentrations was in the order of seeds > whole plant > stem. At a concentration of 1.00 g/L, where the DPPH free radical scavenging rate of seed total flavonoid extract exceeded that of VC and BHT. Correlation analysis indicated a positive correlation between the content of total flavonoid extracts and the scavenging rates of different free radicals, as well as the FRAP. The study demonstrates that the developed HPLC method is simple and provides accurate results, suitable for the determination of flavonoid content in sweet sorghum. The total flavonoid extracts from different parts of sweet sorghum exhibit antioxidant activity. [ABSTRACT FROM AUTHOR] |
