Molecular detection and whole genome sequencing of Pectobacterium and Dickeya, the casual agents of soft rot disease in lettuce.

Abstrakt: Lettuce plants displaying vein discoloration, wilting, soft rot at the base and emitting foul odour were observed at several major lettuce growing areas including Orange-grove and Lopinot in the North, Cunupia in Central and Princes Town in the South of Trinidad between 2017 and 2019. An assessment of disease incidence in four lettuce farms revealed the incidence ranged between 5 and 20%. An isolation of pathogens associated with symptoms of basal rot resulted in similar colonies developing on nutrient agar plates. The use of genus specific primers and whole genome sequencing proved the identity of the organisms. Two pathogens, viz. Pectobacterium brasiliense and Dickeya chrysanthemi were identified as the causal agents for the soft rot disease of lettuce. Genes associated with virulence and pathogenicity, motility, biofilm formation, stress resistance and defense (SrfB, SrfC, Svx/AvrXca, Type IV pilus twitching motility protein PilT, biofilm peroxide resistance protein bsmA, organic hydroperoxide resistance protein, glyoxalase/bleomycin resistance/dioxygenas, arsenic resistance protein, acid resistance repetitive basic protein Asr) were identified. We report for the first time, the full genome sequence and soft rot disease causing agents of lettuce in Trinidad. [ABSTRACT FROM AUTHOR]
