Alginate Lyase Enzyme from Paenibacillus lautus as Potential Candidate for Depolymerization of Alginate to Alginate Oligosaccharides.

Abstrakt: Alginate constitutes the major structural compound of the cell wall and intercellular matrix in brown seaweeds and widely used in the food, pharmaceutical and cosmetic industries. Biological depolymerization of alginate is catalyzed by alginate lyases (EC 4.2.2.3) through a ß-elimination reaction. This enzyme cleaves the molecular chain of alginate and create unsaturated uronic acid at the new non-reducing end. The end product is a mixture of short oligosaccharides, which have been shown to be bioactive and to have antitumour and antivirus activities. These oligosaccharides also have applications in food, pharmaceutical and agriculture industries but limited availability of options for its microbial production necessitated the need for isolation of new microorganisms. In the present study an alginate lyase producing bacterial strain Paenibacillus lautus was isolated and its potential in the formation of alginate oligosaccharides from alginate is assessed and some parameters responsible for maximum production of enzyme were optimized.
