Nepenthes gracilis is a carnivorous plant species with leaves consisting of a photosynthetic organ (lamina) and a pitcher trap. Both organs contain chlorophylls and carotenoids. These pigments absorb light energy for photosynthesis. Studies on both pigments content are essential for understanding the cost-benefit of carnivory in Nepenthes. Fresh samples for pigment extraction from Nepenthes collected directly at their natural habitat can limit the accuracy of both pigment measurements. Hence, this study aimed to assess and determine the differences in chlorophylls and total carotenoids content in fresh and dried laminas, as well as to find the extracting solvent for the highest chlorophylls and total carotenoids concentration. Pitcher plant leaves (fresh and dried) were extracted with four solvents (acetone, ethanol, DMSO, and methanol) and four incubation treatments (0,24,48,72 hours). A spectrophotometry analysis was used to determine chlorophyll a, b, and total carotenoids content. Results showed that chlorophylls content was at the same levels in both fresh and dried leaves. Different extracting solvents resulted in different chlorophyll concentrations in fresh and dried leaves. Fresh leaf samples extracted using DMSO and dried leaf samples extracted using methanol both had the highest chlorophyll content. Differences in the extraction time influenced the chlorophyll b concentration. The highest chlorophyll b concentration was detected after 72 h extraction using DMSO. In contrast, methanol extraction of chlorophyll b in 0 h could result in the highest concentration. These results are essential in understanding and designing better sampling protocols for preserving materials used for compound analysis from wild plants. [ABSTRACT FROM AUTHOR] |
