A review of solid state mechanochemistry for drug synthesis and modification.

Abstrakt: Different aspects of the use of mechanochemistry in the study of pharmaceutical materials are discussed, including the use of mechanical activation to modify the physical and chemical properties of drugs and the use of mechanochemical methods for solid-state synthesis of drugs. During grinding and tabulation, mechanochemical variables must be considered. A branch of solid-state chemistry known as mechanical chemistry studies processes that can occur in solids when mechanical energy is applied. The energy is provided by high-energy milling. Mechanochemical approaches produce a variety of surface and bulk defects in treated solids, which intensifies the course of processes. A milling process accumulates energy which can be used in a subsequent process. Mechanochemistry has the potential to be utilized in cases where the economic demand, the high temperature as well as wet processes may need to be considered. It will be demonstrated how a mechanochemical approach can be applied to such widely diverse fields as extractive metallurgy, materials engineering, and medicine. The aim of this review is to show how mechanochemical techniques have advanced the development of new solid forms as well as a structural understanding of these forms, plus the use of supramolecular strategies to create pharmaceutical solid forms.
