Abstrakt: The sperm-specific Na+/H+ exchanger SLC9C1 stands out by its unique tripartite domain composition. It is the first known secondary-active transporter which is directly activated by membrane voltage via a voltage-sensing domain (VSD). Our cryo-EM structures of sea urchin SLC9C1 reveal the domain arrangement and new structural coupling elements, allowing us to propose a mechanism for a voltage-activated alternative access mechanism.
