Uncaria gambir is one of the widely used traditional medicine in Indonesia, but it is still, unclear in its molecular identification. DNA barcoding provides an advanced method in plant identification. In this study rbcL gene was used as barcode DNA to discriminate Uncaria gambir from another Uncaria species. The genomic DNA of Four Uncaria gambir plants was extracted and amplified by rbcL specific primer, MEGA-X was used for species distance analysis, and a Maximum Likelihood phylogenetic tree was also constructed. The rbcL barcode gives 50 % amplification efficiency and no barcoding gap intraspecific and interspecific value. Phylogenetic tree analysis showed Uncaria gambir closely related to Uncaria macrophylla. [ABSTRACT FROM AUTHOR] |
