Localization of Telomere-Binding Protein TRF2 in Spermatogenic Cells of Hibernating Frogs Rana temporaria.

Abstrakt: Telomeric DNA is associated with a protein complex, which is called "shelterin." There are data indicating that the functions of the shelterin complex are wider than just protecting chromosome ends. There are examples of unusual distribution of the shelterin component, telomere-binding TRF2 protein, in mouse and common frog Rana temporaria oocytes and mouse spermatozoa. In this work, the distribution of TRF2 in the cells of spermatogenic row of the common frog was studied. To confirm the specificity of antibodies to the grass frog TRF2, Western-blot analysis of testis preparations was used. Antibodies selected by immunoblotting were used to determine the localization of TRF2 in spermatogonia and spermatozoa using immunoelectron microscopy. It was demonstrated that TRF2 in spermatogonia is evenly distributed throughout the nucleus. The presence of TRF2 was detected in the cytoplasm of spermatogonia in electron-dense structures. In spermatozoa, some of the TRF2 is located in the acrosome, proving the universality of the phenomenon for vertebrates. [ABSTRACT FROM AUTHOR]
