This study compared the radiosensitivity of chromosomes 1 and 4 in peripheral blood lymphocytes obtained from a single donor living in a region with high concentrations of environmental radon. The break locations in these chromosomes were also examined. Blood samples obtained from a single donor living in an area with high concentrations of environmental radon were irradiated with 2 Gy of γ-rays. Peripheral blood mononuclear cells were isolated and cultured for 48 hours. Chromosomes 1 and 4 were painted using red and green fluorescent probes, and chromosomal aberrations were scored from 150 images of metaphase cells. Breakpoint analysis was also performed to identify the distribution of break positions in each chromosome. Chromosome 1 was more frequently involved in both reciprocal and non-reciprocal translocations than chromosome 4. Chromosome 1 also showed a higher frequency of dicentric chromosomes and acentric fragments than chromosome 4. Fewer break positions were observed near the ends of the arms on chromosome 1. Similar findings were observed for chromosome 4. More breaks were observed in the middle region of each arm of chromosome 1 than were observed for chromosome 4. The results of our study supported the assumption that the radiation sensitivity of chromosomes is correlated with the chromosomal DNA content or chromosome size. Our study results also supported the non-random distribution of chromosome breaks. In the examined individual exposed to high concentrations of environmental radon, chromosome 1 was more sensitive to radiation than chromosome 4. The distribution of break locations was not random in either chromosome. More investigations should be performed using different radiation doses, and the existence of a gender effect should be examined to verify the findings of this study [ABSTRACT FROM AUTHOR] |
