Responding to the decreasing herpetofauna diversity species in potential locations in Malang, Indonesia, research on Amphibia should be carried out, especially in the Genus Microhyla which often observed in cryptic species. This study aims to evaluate and contribute to the database for taxonomic studies of animals, especially frogs of the genus Microhyla. In total, 10 samples were used, consisting of 9 Microhyla orientalis and 1 Microhyla palmipes. 16S rRNA gene was amplified using 16L2 forward primer and 16H1 reverse primer. 22 additional sequences from GenBank were used for phylogenetic reconstruction (including outgroup) with Bayesian Inference. Phylogenetic reconstruction produced a well- resolved tree with five clades and well-supported node branches. M. orientalis is located as the sister taxa of M. malang. While, M. palmipes from Malang (East Java) are closely related to M. palmipes from Bali, compared to the M palmipes from Sumatra. Intraspecific genetic distance analyses of M. palmipes show a quite big distance so that we assume there is a chance to be novel species. Future studies are suggested to involve more samples to evaluate the taxonomy of this species. [ABSTRACT FROM AUTHOR] |
