DNA Sequencing of blaSHV Genes among Uropathogenic Klebsiella pneumoniae Harboring blaCTX-M-15.

Abstrakt: Introduction: The spread of antimicrobial resistance in ESBLs Klebsiella pneumoniae strains usually makes nosocomial infection treatment difficult. Mostly, ESBLs found in this pathogens carry the TEM and SHV, this study was designed to detect the SHV variant among two K. pneumoniae isolates harboring blaCTX-M-1s and methods: Two uropathogenic Klebsiella pneumoniae strains were identified, and by using the recommended methods cephalosporins susceptibility was detected. Universal primer for blaSHVand then products were subjected to sequencing. Results: The two isolates showed highly resistant to all tested cephalosporins different generations, at which the MIC results for cefradine, cefuroxime and cefixime were > 128 μg/ml, 128 μg/ml, 32 μg/ml and > 128 μg/ml, > 128 μg/ml, > 128 μg/ml for strains K42 and Kp7 respectively. The sequencing of amplicons from the blaSHV group showed high identity with different known blaSHV like variants. Conclusion: The occurrence of blaSHV like genes among two Egyptian ESBL producing Klebsiella pneumoniae strains was demonstrated in this study. High similarity with different other global genes in the gene bank documented in NCBI was obtained. Therefore, further studies on a large blaSHV carrier of Klebsiella pneumoniae are in need to detect the prevalence of these genes. [ABSTRACT FROM AUTHOR]
