Anti-breast Cancer Activity of Co(II) Complex by Inhibiting Cell Viability and Stimulating Cell Apoptosis.

Abstrakt: In this research, a novel low-dimensional Co(II) coordination compound, namely, Co(H2pimdc)2(phen) (1), was prepared through the reaction of Co(NO3)2·6H2O and 2-propyl-imidazole-4,5-dicarboxylic acid (H3pimdc) under solvothermal reaction conditions using 1,10-phenanthroline (phen) as the co-ligand. Serial experiments were conducted for the biological activation assessment, and the related mechanism was explored as well. Firstly, the inhibitory activity of the new compound on the viability of the breast cancer cells was evaluated by the Cell Counting Kit-8 assay. The percentage of the apoptotic breast cancer cells after compound treatment was measured with Annexin V-FITC Apoptosis detection. Then, the western blotting assay was performed and the expression of the BCL-2 protein was measured. Finally, the real expression of the angiogenesis genes was measured with real time reverse transcription-polymerase chain reaction. [ABSTRACT FROM AUTHOR]
