The Effect of Mitomycin C on Antigenic and Mitogenic Stimulation of Mouse Lymphocytes In Vitro.

Abstrakt: The effect of mitomycin C on DNA synthesis and plaque formation in mouse spleen cells was studied in vitro, with special reference to the synergistic effect observed between some antigens and mitogens on lymphocyte stimulation. Mitomycin c inhibited DNA synthesis in doses that did not cause an extensive cell death. I blocked almost completely a primary plaque-forming cell response to sheep erythrocytes (SRC) and, to a great extent, PPD-induced antibody synthesis detected against SRC. The synergistic effect earlier observed in vitro between SRC and PPD was not abolished in cultures where replication of DNA was depressed. Simultaneous stimulation with SRC and PPD in cultures treated with mitomycin C allows an estimation of the number of antigen-sensitive cells to SRC that are stimulated to antibody synthesis without prior cell division.
