To investigate the expression and significance of pyruvate carboxylase (PC) for astrocytes cells, we evaluated the effect of the PC inhibition on survival of cultured human astrocytes cells and the expression of PC in cultured astrocytes cells as well as among human brain cortex samples by immunoprobing methods. Our experimental results showed that the combination of andrographolide and 6-shogaol exhibited the most prominent NO inhibitory effect in lipopolysaccharide (LPS)-activated microglia N11 cells with IC SB 50 sb value of 6.10 M. This combination continued to show a promising effect in LPS activated tri-culture system (microglia N11, endothelial bEnd3 cells, and neuron N2A cells) that reduced NO production and enhanced neuronal survival.
