This article discusses about an examination of major histocompatibility complex glycoprotein (MHC) restriction in the context of a minimal clonal abortion model for self tolerance. The fact that T-cell responses to non-self (virus) are always constrained by association with MHC has general implications for the theory of immunological tolerance. The tolerance problem may no longer be seen simply as a need to distinguish between two opposites, self and non-self. The central question now concerns the nature of the distinction between self and self plus other (self+x). The physiological mechanisms underlying MHC restriction and self tolerance must be considered concurrently. |
