Abstrakt: This article reports a study measuring apparent kinetic parameters for NADPH involvement in hepatic drug metabolism with microsomes from phenobarbital pre-treated rats. The study also examines the influence of both NADH and a structural analogue 1,4,5,6-tetrahydronicotinamide adenine dinucleotide on these kinetic parameters in an attempt to further clarify the control of hepatic drug metabolism. In the present work with phenobarbital pre-treated animals where the specific activity of NADPH-cytochrome P-450 reductase is significantly higher there is no stimulation of this transfer from NADPH in the presence of NADH even though a reduction in NADPH values suggest that a similar interaction is still occurring between NADH and cytochrome P-450-substrate complex. This failure of added NADH to stimulate the rate of cytochrome P-450 reduction in the phenobarbital induced microsomes indicates a fundamental difference between induced and non-induced hepatic microsomes.
