DEATH OF CELLS BY APOPTOSIS FOLLOWING ATTACHMENT OF SPECIFICALLY ALLERGIZED LYMPHOCYTES IN VITRO.

Abstrakt: This article cites findings of a light and electron microscopic study of death of cultured tumour cells incubated with splenic lymphocytes harvested from animals previously immunized against the tumour. Special attention has been paid to events that follow lymphocyte attachment, which is widely considered to be an essential preliminary step in the killing process. In this study, all cell cultures and killing experiments were performed at 37 degree centigrade in a water-saturated atmosphere containing 5% carbon dioxide. Mastocytoma cells were grown in RPMI 1640 culture medium, supplemented with 10% heat-inactivated foetal calf serum. The reaction in individual tubes was monitored at various times up to 6 hours using 3 procedures. It was noticed that apoptosis occurred spontaneously in the control mastocytoma cultures, as it does in a variety of neoplasms growing in vivo.
