Optimization of immobilizing conditions for inulosucrase with sodium alginate by response surface methodology.

Abstrakt: The experiment was to study the immobilizing condition of inulosucrase. The enzymatic activity of inulosucrase as response value, the immobilizing conditions were optimized by single factor and response surface test. The results showed that optimal conditions of immobilization were as follows: Sodium alginate concentration 2.8%, calcium chloride concentration 3.8%, immobilization time 3.0 h, glutaric dialdehyde concentration 0.035% and cross-linking time 30 min. Using these conditions, the enzymatic activity of immobilized inulosucrase was 78.82 U/g. The experiment indicates that immobilized inulin sucrase has better thermal stability and pH value stability than free enzyme. After 10 cycles, the immobilized enzyme activity still maintained 61.15% of the original enzyme activity. [ABSTRACT FROM AUTHOR]
