Phalaenopsis is one of the most popular orchid plants in the world and has high demand in the global market. However, this orchid is difficult to propagate vegetatively. Hence, propagation through tissue culture is desirable to produce a uniform and mass quantity of plantlets. This study aimed to develop in vitro shoot regeneration of Phalaenopsis Sunny Yellow from flower stalk nodes. The best treatment for surface sterilization of the explants were by addition of 1% Benomyl and 10% sodium hypochlorite solution which produces 80% survival rate. The explants were then cultured on Murashige and Skoog (MS) media with different combinations of hormones; 1- Naphthaleneacetic acid (NAA), Thidiazuron (TDZ), Benzyl Aminopurine (BAP) and Indole Acetic acid (IAA). Results showed that MS media supplemented with 1mg/L NAA and 1mg/L TDZ produced new shoots with an average number of 8±1.4 shoots per explant and 5.7±1.31 cm shoot length, respectively. This protocol provides information on induction of in vitro culture of Phalaenopsis Sunny Yellow for mass propagation via tissue culture method. [ABSTRACT FROM AUTHOR] |
