When ultrathin sections of normal epidermis were stained with uranyl acetate, but especially when the tissue blocks in addition were prestained with uranyl acetate while being dehydrated, each keratinocyte showed a densely stained continuous layer peripheral to the light mid-layer of the trilaminar plasma membrane. Within desmosomes this dense layer was quite regular in thickness and corresponded to the intermediate dense layer, as described by Odland (1958). Outside of desmosomes, however, the dense layer was very irregular in thickness, indicating that this layer was not identical with the outer leaflet of the plasma membrane. In pemphigus acantholysis the dense layer within desmosomes was largely dissolved and thus the much thinner outer leaflet of the plasma membrane was revealed. It is concluded that the dense layer surrounding each keratinocyte, including the intermediate dense layer of Odland, Is composed of the outer leaflet of the plasma membrane and a variable amount of intercellular substance condensed on it as `a cell surface coat.
