Dofetilide is a class III antiarrhythmic agent that is widely used for chronic atrial fibrillation (CAF). In this study, the hypothesis was tested that dofetilide effects on the electrical activity in cardiomyocytes may differ depending on the age, particularly under CAF condition. Populations of Courtemanche98 ionic models were built to simulate effects of CAF on the action potential (AP) generation in the adult and ageing atrial cardiomyocytes and to unravel age-dependence of AP response to dofetilide. The results showed that dofetilide effects on AP in virtual populations of normal atrial cardiomyocytes essentially declined with age. The AP response to dofetilide was less expressed in cardiomyocytes under CAF in comparison with the normal cardiomyocytes and was almost age-independent. [ABSTRACT FROM AUTHOR] |
