In this study, evaluated the potential of rifampin production and characterize by used HPLC (High-performance liquid chromatography) analysis pattern of actinomycetes. A total of 18 samples from Beach Ridges Interspersed with Swales (BRIS) soils were collected from a different depth of the earth from Setiu district, Terengganu by using the spread plate technique on different agars. Seven genus of rare actinomycetes strains obtained and have been designated by 16s rRNA, GenBank, and sequence similarity by phylogenetic analysis. Rifampin was taken up for qualitative and quantitative HPLC analysis as standard to compare with cruds extract depends on rotation time. Following the extraction of the secondary metabolite, the HPLC analysis was carried out for the characterization of various extracts. Considering the coordinate analysis of the HPLC pattern, isolates AA12, AA13, and AA17 might be a potential producer of Rifampin and isolates AA8, AA78, AA83, and AA141 can't produce the Rifampin, but this isolates might be producing to other antibiotic compounds. The results for secondary screening for crude extracts against six pathogenic microbial showing inhibition zone in different proportions. These results highlight the importance of actinomycetes isolates for the production of antibiotic compounds. Where previous studies have proven that many compounds found in actinomycetes exhibit antimicrobial activity, analgesic, anti-inflammatory, and antioxidant properties. [ABSTRACT FROM AUTHOR] |
