Previous studies have demonstrated the effectiveness of Lysate-PRF as a culture medium on proliferation and differentiation of human dental pulp stem cells (hDPSCs). Therefore, this study was conducted to continue those previous studies with the primary aim of analyzing the effects of lysate platelet-rich fibrin (PRF) culture medium on VEGF-A expression of hDPSCs. Moreover, determination of the optimum concentration lysate PRF in inducing VEGF-A expression of hDPSCs was also tested. HDSPCs were isolated from premolar teeth extracted for orthodontic purposes from healthy donors. After 24-hour serum starvation, hDPSCs were cultured in Dubelco's Modified Eagle Medium (DMEM) as a negative control, Lysate-PRF diluted with DMEM with concentrations of 1%, 5%, and 25%, and DMEM supplemented with 10% FBS as a positive control. HDPSCs were planted into 96-well plates at a concentration of 2x104 cells/mL and each group was triplicated under the same treatments. Each group was tested for VEGF-A expression in a 24-hour evaluation using ELISA. All Lysate-PRF groups showed escalation of VEGF-A expression of hDPSCs. Compared to 5% and 25% Lysate-PRF concentrations, 1% Lysate-PRF was the optimum concentration for inducing VEGF-A expression of hDPSCs. 1% Lysate-PRF's ability to stimulate VEGF-A expression of hDPSCs was not statistically significant compared to 10% FBS. A 1% Lysate-PRF concentration has equal effectiveness in inducing VEGF-A expression of hDPSCs when compared to 10% FBS. [ABSTRACT FROM AUTHOR] |
