The main aim of this study was to determine the effect of dietary B-glucan extracted from Shiitake Mashroom (Lentinan) as detoxifier to Fumonisins B1 in mice. This study included the followings: The first step was carried out to determine the percentage of Fusarium verticillioides in maize of poultry diet by collection 50 feed samples from different poulty farms and markets in Baghdad province (Abu-Ghrab, Al-Mahmodia, Al-Madain, Al-Rashydia) during (Novmber 2019 to March 2020). These samples were cultured on (potato-dextrose agar) for isolation and identification of F. verticillioides. The result of this part showed (86%) of maize samples were contaminated with fungi and particularly (13.95%) were contaminated with F. verticillioides. The second part of this study included extraction of FB1 from 6 isolates of F. verticillioides by culturing on rice medium and measuring the FB1 concentration by Enzyme Linked Immune Sorbent Assay (ELISA), where found FB1 in different concentrations (9.8, 8.9, 6.4, 6.2, 4.4 and 4.1 ppm). The highest concentration was used in experimental study which included using 40 albino mice divided into 4 groups with 10 animals into each group. The 1st group (control -ve) was fed on normal feed without mycotoxin. The 2nd group (FB1) treated with 100µg/kg bw of FB1 orally for 2 weeks. The 3rd group (FB1 with LNT) was treated as the 2nd group and treated with 3mg/mouse of LNT orally for 7 days. The 4th group (LNT) was treated with 3mg/mouse of LNT orally for 7days. And the body weight was measured at the bignning and end period of experiment. The blood samples were collected by heart puncture at the end of experiment to obtain of serum that used for evaluation of liver functions. All animals were scarified and some internal organs like (liver, kidney, intestine, spleen and lung) used for studying the histopathological changes. The results showed that the FB1 caused significant decrease (P≤0.05) of body weight of mice in 2 nd group, while mean body weight increased significantly (P≤0.05) of LNT group due to the effect of LNT an effective biostimulant and no significant differences between 3rd group and 1st group due to lentinan effect as detoxifier to FB1. In concerning of detection of liver enzymes activites (ALT, ALP and AST), the result showed significantly increased (P≤0.05) in 2 nd group and 3 rd group. While, there are no significant differences between 4 th group and 1 st group. Finally, the histopathological changes of all organs of the 2 nd group were appeared and characterized by apoptosis, necrosis, degeneration and atrophy. While, in other groups (3rd and 4th) there were no changes was observed. Therefore, the Lentinan can be used in the dietry feed of animal's in order to limit and avoid the effects of FB1 mycotoxin and enhancing the immunity in the animal's body [ABSTRACT FROM AUTHOR] |
