SEQUENCE CHANGES OF E CADHERIN CDH1 GENE ENCODING THE TRANS MEMBRANE DOMAIN IN INFILTRATING DUCTAL CARCINOMA OF THE BREAST.

Abstrakt: The mutations of the cell adhesion protein E- Cadherin is predicted to play a major role in the development of epithelial cancers. Studies had shown a strong association of E- cadherin mutations and breast cancer . The present study was aimed at studying the exonic sequences, exon 13 and 14 of E-cadherin in ductal carcinoma samples of the breast and the sequences were compared with the same exonic sequences from normal samples using the NCBI BLAST tool. The functional implication of the observed changes was studied using the NCBI ORF Reading frame finder. The exonic region 13,14 encodes the trans membrane domain of the E-cadherin protein which has a major role in the adhesive function of the protein. We observed four sequence changes in the exonic regions namely deletion of A at two locations ; an insertion of A in other location and a transversion of C to T in the tumour sequences of South Indian ethnic population . Upon comparison of the protein sequence of the tumor and normal samples two changes were observed namely a deletion of leucine residue and a replacement with glutamine instead of histidine was observed with the tumor protein sequences .
