The article presents information on Keratinocyte Grafts and Skin Equivalents. Advances in tissue culture methods permit the growth of these individual cells from small skin biopsy specimens. This brought to the attention of dermatologists and burns surgeons the possibility of expanding tissue by using cultures in skin grafting: keratinocyte grafts or cultured epithelial grafts. Initial attempts to culture skin in vitro involved organ cultures and later explant cultures in which a skin sample was incubated in a dish with culture medium. The epidermis grew down the side of the explant and migrated outward to produce epithelial layers; however, the proliferation of epidermal cells was insufficient to produce the amounts necessary for grafting and fibroblast overgrowth was a problem. |
