Nucleotide variation is important information for basic strategy in conservation programme of animals. NADH dehydrogenase subunit-2 (ND2) gene is one of the protein codings of mitochondrial DNA of animal. This study informed nucleotide variation between two species of Lorikeet (genus Trichoglossus; T. flavoviridis and T. ornatus) birds from Sulawesi, Indonesia. DNA was extracted from blood samples of 10 birds; consisted of three of T. flavoviridis birds and seven of T. ornatus birds. A single DNA fragment (1 100 base pairs) of ND2 gene of each bird was successfully amplified using a pair of nucleotide primer in the PCR (Polymerase Chain Reaction) and then be sequenced. DNA sequence data (1 011 base pairs) from all birds were analysed using Mega 5 Version and DNASP4 software. There were no stop codons in the all DNA sequence data, indicated that DNA fragments target are a protein-coding gene and comprised of 337 amino acids. Between the birds there were 80 polymorphic (segregating); consisted of 50 Singleton variable sites and 30 parsimony informative sites. 10 sequence haplotypes (HT1-HT10) were found with haplotype diversity was 1.000 ± 0.045. Nucleotide diversity (Pi) was 0.0279 ± 0.0053. Mean Genetic distances between T. flavoviridis and T. ornatus was 0.046 ± 0.006, within T. flavoviridis was 0.005 ± 0.004; and within T.ornatus was 0.0072 ± 0.002.
